Variables
Description
What is this formula?
The Therapeutic Window Ratio (TWR) compares the minimum concentration at which toxicity begins to appear with the minimum concentration required to achieve a therapeutic effect.
When to use it
Use this formula when evaluating drugs whose efficacy and toxicity are best described using plasma concentrations rather than administered doses.
Example
A drug has a minimum effective concentration of 2 mg/L and a minimum toxic concentration of 12 mg/L.
Data:
MEC = 2 mg/L
MTC = 12 mg/L
Formula:
TWR = MTC / MEC
Substitution:
TWR = 12 / 2
Result:
TWR = 6
Interpretation:
The toxic concentration threshold is six times higher than the minimum concentration required for therapeutic efficacy.
Applications
- Clinical pharmacology
- Therapeutic drug monitoring
- Drug development
- Dose optimization
- Pharmacokinetic studies
Note
The Therapeutic Window Ratio is a simplified measure of drug safety based on concentration thresholds. In clinical practice, therapeutic windows are often represented as concentration ranges rather than a single ratio. The exact values of MEC and MTC may vary among patients due to age, genetics, organ function, drug interactions, and disease state. Alternative definitions using doses instead of concentrations may also be found in the literature.
